Antiviral tests

tekFABRIK™ has been tested against non-enveloped virus, bacteriophage, enveloped virus such as common respiratory viruses (influenza and respiratory syncytial virus) and clinical isolates of SARS-CoV-2, the COVID-19 causing virus. Tests were performed in four independent laboratories using a modification of ISO 18184 "Textiles - Determination of Antiviral Activity of Textile Products."

Killing of SARS-CoV-2

A sensitive RT-PCR assay was used to quantitate virus inactivation by tekFABRIK™ after incubation of a clinical isolate of SARS-CoV-2 . The assay was performed at a national testing laboratory in India (RGCB, Thiruvananthapuram, India). The test amplified two viral genes the envelop (E) gene and spike protein (S) of the virus. Results show that 99% of the virus is killed after 30 min on tekFABRIK™.


Killing of Influenza Virus A/WSN/H1N1

The virus has been engineered to express GFP gene for monitoring the virus on infection of host cells.

Results show that MDCK cells infected with virus pretreated on tekFABRIK™ show no GFP fluorescence virus spotted on control cloth were live and infected MDCK cells as exhibited GFP fluorescence, This confirmed that chemically modified TekFABRIK™ has viricidal properties against H1N1.

Killing of Respiratory Syncytial Virus (RSV)

tekFABRIK™ was tested for its viricidal activity against RSV in a US academic laboratory that has studied RSV for many years and has developed a sensitive luciferase-based assay to monitor RSV infection. The genetically engineered RSV (rA2-Rluc) was incubated with tekFABRIK™ according to a modified ISO 18184 procedure. At the indicated times, the virus was recovered from the cloth and virus infectivity measured by incubation with A549 cells, a human lung epithelial line ( (ATCC, CCL-185). tekFABRIK™ had high viricidal activity against RSV with 97% reduction in viral infectivity after 5 min as determined in three independent experiments. Killing of bacteriophage P1

Phages are non- enveloped virus that infect E.coli and are routinely used a model to test a for viricidal activity. Also, bacteriophages can be studied in a standard research lab with biosafety level 1 Bacteriophage was spotted on tekFABRIK™ / control fabric for different time periods and then plated by Agar overlay assay. After 24 h, the plates were observed for plaques. Alternatively, the E.coli infected with phages is directly plated on LB agar to determine the viricidal activity.

Anti-bacterial activity of tekFABRIK™

TekFABRIK™ has been extensively studied and shown to have broad – spectrum anti- bacterial properties encompassing both gram positive and gram negative bacteria. For example, two strains commonly associated with hospital acquired infections, Staphylococcus aureus (gram positive) and Klebsiella pneumoniae (gram negative) were tested and the results are shown below. An inoculum of 1-2 x105 in PBS was spotted on TekFABRIK™ of control cloth and incubated for the indicated times. The bacterial cells were harvested from both the TekFABRIK™ and control cloth and plated on Tryptic soy agar and incubated overnight at 37°C. The number of colony forming units (cfu) was measured and converted to % kill as a function of time using the formula:

(cfu/mL (control cloth) – cfu /mL(tekFABRIK)/Total cells spotted) x 100 = % Kill
